There seems to be a problem with NextSTART not saving this sound when it's configured in Prefs. However, adding this line in mytheme.wst seems to work, but only after restarting Windows 10:

FxSound11=C:\Users\Public\Documents\Winstep\Themes\MyTheme\Detach.wav

Thought this would seem the likely candidate - is it correct?

Trying to add the new sound to some recent themes, so thought I'd check before attempting any more...

The detach sub menu sound is being properly saved.

The problem is that all sounds are first reset (i.e.; set to nothing) when you switch to a different theme, unless you have 'Prevent themes from changing your current sound scheme' enabled in the Sounds & Animations tab of NextSTART Preferences.

The only bug here is that the default 'Detach Sub-Menu' and 'Take Picture' sounds should remain but they weren't.

I've tried it again in two different themes. This time without switching themes. Just restarting Winstep, and the Detach sound's gone. Quit Winstep and restart Windows and Winstep, also not saved. Add in the line in MyTheme.wst from my first post and restart Windows though does do it. :)
